Merkmale / Symptome
|
Erkrankung der Glykogenspeicherung in der Muskulatur (übermässige Glykogenspeicherung = Thesaurismose). Phänotypische Variabilität (Genexpression spielt eine Rolle in der Ausprägung der Krankheit, Teixeira R et al 2015). Hauptsymptome wie beim Kreuzschlag/Kreuzverschlag, erfolgen jedoch 10 bis 20 Minuten nach leichter Arbeit, manchmal gar diskret: Muskelsteifheit, Bewegungsunlust bis hin zur absoluten Unfähigkeit sich zu bewegen, Schmerzen, Lahmheiten, breitbeinige Position; erhöhte Laborwerte (Muskelenzyme), in schweren Fällen Myoglobinurie. Meist leichtfuttrige Pferde sind betroffen. Die Auswirkung scheint bei den Homozygoten gravierender als bei den Heterozygoten zu sein. Doppelgenträger (PSSM and MH) erkranken schwerer (McCue ME et al, 2009b). |

Erblich bedingte Einschränkung einer Organfunktion (Stoffwechsel der Muskeln); Bewegungsstörung, Schmerzen. Schäden, die zu Funktionsausfällen führen, die den Allgemeinzustand beeinträchtigen Massgebende Kriterien für die Beurteilung der Belastungen sind insbesondere:

Eltern
DNA-Test; gezielte Anpaarungen; Anpassung der Zuchtstrategie (Mutationsträger sollen in der Zucht nicht eingesetzt werden) Betroffene Nachkommen
Diagnose (DNA-Test); befallene Tiere sollten in der Zucht nicht eingesetzt werden; palliative Massnahmen: abklären, ob das Pferd überhaupt Kraftfutter benötigt, ausbalancierte kohlenhydratarme Diät (ausreichend Heu als Grundfutter; bei intensiverer Arbeit: Kohlenhydrate in der Ration zum Teil durch Speiseöl ersetzen, Mineralstoffmischung mit Vitamin E und Selen); Anpassung der Nutzungs- und Haltungsform: regelmässige und tägliche Bewegung, Paddock- und Weidehaltung. |

|
Wissenschaftler gehen davon aus, dass dieses Gen bereits vor 1200-1500 Jahren mutiert ist, also vor der Entstehung moderner Rassen (McCue ME et al, 2008b). Eine Myopathie PSSM Type 2 (GYS1-negative mit PSSM: PSSM2) wird beim Quarter Horse und Warmblüter beschrieben (McCue ME et al, 2009a; Teixeira R et al, 2014). GWAS: SNP in ECA3 (nicht signifikant nach einer Bereinigung der Populationsstruktur), ECA11, ECA18 (falsch positiv) und ECA27 (Fritz KL et al, 2012b; Teixeira R 2015). Keine eindeutige kausale Mutation wurde identifiziert. Hypothesen: Mutation im PFAS-Gen (ECA11), Populationsstruktur (schwaches Kopplungsungleichgewicht beim Quarter Horse), PSSM2 ist eine polygenetische Krankheit oder PSSM2 ist ein Sammelbegriff für verschiedene Phänotypen (Teixeira R 2015). Die Diagnose ist bisher nur über Muskelbiopsien möglich (McCue ME et al, Webseite). PSSM gehört zum Krankheitsbild der belastungsinduzierten Myopathien (oft idiopathisch und sehr unterschiedliche Symptomatik: sporadisch, wiederkehrend; leichtgradige bis schwere schmerzhafte Muskelkrämpfe während oder nach der Arbeit): Kreuzschlag, Exertional rhabdomyolysis (ER), Tying up Syndrom (Syn. Recurrent Exertional Rhabdomyolysis, RER; wiederkehrende belastungsinduzierte Myopathie, s. University of Minnesota). Nur bei einer bestimmten Gruppe von Pferden liegt eine PSSM-Erkrankung vor. Mögliche Genregionen für andere belastungsbedingte Myopathien bei Vollblüter: ECA12 und ECA20 (Tozaki et al. 2010; Region 29–42 Mb ECA16, Fritz KL et al, 2012a); ECA14 (Norton E et al, 2014) Folgende erbbedingte Erkrankungen der Skelettmuskulatur wurden beschrieben:
